Combination of commercially available molecular assays and culture based methods in diagnosis of tuberculosis and drug resistant tuberculosis BJM

ABSTRACT Early diagnosis of tuberculosis is of major clinical importance. Among 4733 clinical specimens collected from 3363 patients and subjected to Ziehl-Neelsen microscopy, 4109 were inoculated onto Löwenstein-Jensen slants and 3139 in Bactec/9000MB. Polymerase Chain Reaction (PCR) was performed in 3139 specimens, whereas, a genotypic assay was directly applied in 93 Mycobacterium tuberculosis complex PCR-positive for isoniazid and rifampicin resistance detection specimens (GenoType MTBDRplus). Comparative analysis of cultural isolation and PCR based assay for detection of Campylobacter jejuni in food and faecal samples BJM

In the present study, the efficacy of polymerase chain reaction (PCR) based on mapA gene of C. jejuni was tested for detection of Campylobacter jejuni in naturally infected as well as spiked faecal and food samples of human and animal origin. Simultaneously, all the samples were subjected to the cultural isolation of organism and biochemical characterization. The positive samples resulted in the amplification of a DNA fragment of size ~589 bp in PCR assay whereas the absence of such amplicon in DNA extracted from E. coli, Listeria, Salmonella and Staphylococcus confirmed the specificity of the primers. The objective of the present study was to determine the sperm enrichment with X-bearing spermatozoa, after one centrifugation in a Percoll or OptiPrep continuous density gradient, using quantitative real-time polymerase chain reaction (qPCR) of sperm DNA and resultant in vitro-produced bovine embryos by PCR. Comparison of DNA-extraction methods and Selective Enrichment broths on the detection of Salmonella Typhimurium in swine feces by polymerase chain reaction (PCR) BJM

The aim of this study was to compare different DNA-extraction methods and selective enrichment broths for their effectiveness to detect Salmonella Typhimurium in artificially inoculated swine feces samples (100 CFU/g) by polymerase chain reaction. After enrichment in Rappaport-Vassiliadis, selenite cystine or Müller-Kauffmann tetrathionate, aliquots were used for DNA extraction by three different methods: boiling-centrifugation, phenol-chloroform and salting-out. Aliquots of extracted DNA were then used as template in PCR. The selective enrichment broths had no effect on the efficiency of PCR when boiling-centrifugation and salting-out were used. Comparison of phenotypic and molecular tests to identify lactic acid bacteria BJM

Comparison of the specificity of PCR and the histopathological detection of leishmania for the diagnosis of American cutaneous leishmaniasis BJMBR

More precise and rapid diagnostic methods for American cutaneous leishmaniasis (ACL) are necessary because of the growing number of cases observed in Brazil, including the northeastern region of the State of São Paulo. We applied PCR to 54 skin or mucosal biopsies from patients with a clinical and/or laboratory diagnosis of ACL using primers 13A and 13B, with positive results being obtained for 82% of the samples. When the PCR results were compared to those of histopathological leishmania detection, PCR showed superior results with 81.5% sensitivity and 95% CI of 68.0-95.1%. Comparison of three laboratorial tests for diagnosis of canine parvovirus infection Arq. Bras. Med. Vet. Zootec.

The aim of this study was to evaluate the rapid tests currently used for canine parvovirus (CPV) diagnosis: hemagglutination test (HA), enzyme immunoassay (EIA) and polymerase chain reaction (PCR). A total of 112 fecal samples collected from diarrheic puppies up to one year of age were tested. The EIA was able to detect CPV antigen in 44 samples. By HA, 32 samples tested highly positive with titers &gt;128, eight tested weakly positive (titers 32 and 64) and 72 were negative (titers <16). Using PCR, 57 samples were found positive including 13 EIA-negative and 19 HA-negative samples. The best correlation was observed between EIA and PCR (88.4%). Comparison of two slide agglutination serotyping methods and PCR-based capsule typing for the characterization of Haemophilus influenzae serotypes BJM

Ninety-three nasopharyngeal Haemophilus influenzae isolates were serotyped by two slide agglutination methods (SAST 1 and SAST 2) and the results compared with those obtained by capsule type-specific PCR. SAST 1 presented a low correlation with results obtained by PCR (75.2%) while SAST 2 showed a better agreement with the molecular technique results (93.5%). These findings suggest that SAST 2 could be an alternative method for adequate detection of H.influenzae type b.

A avaliação da contaminação ambiental por Bacillus cereus foi realizada em 90 amostras de ar ambiente e em 96 amostras de superfícies de bancadas e de equipamentos, de dois restaurantes institucionais. O microrganismo foi detectado em 84,4% e 44,8% das amostras de ar ambiente e de superfícies, respectivamente. O potencial enterotoxigênico dos isolados foi investigado através da reação da polimerase em cadeia (PCR) para os genes hblA, hblD e hblC (que codificam a hemolisina BL) e para os genes nheA, nheB e nheC (que codificam a enterotoxina não hemolítica - NHE).
